Total synthesis and structural revision of (±)-nidemone

Total synthesis of the revised structure of (±)-nidemone has been accomplished from 6-bromo-2-hydroxybenzaldehyde (5) in either six or eight synthetic steps in 6% and 10% overall yield, respectively, without using any protecting group. The revised structure of nidemone was confirmed by single-crystal X-ray diffraction analysis. Sonogashira coupling, regioselective hydrogenation, and an intramolecular Stetter reaction were the key steps in the synthesis.

Organic & Biomolecular Chemistry (OBC) publishes original and high impact research and reviews in organic chemistry. We welcome research that shows new or significantly improved protocols or methodologies in total synthesis, synthetic methodology or physical and theoretical organic chemistry as well as research that shows a significant advance in the organic chemistry or molecular design aspects of chemical biology, catalysis, supramolecular and macromolecular chemistry, theoretical chemistry, mechanism-oriented physical organic chemistry, medicinal chemistry or natural products.
